摘要:
目的 探讨瑶浴清消方(YYQXF)对急性湿疹小鼠模型的干预治疗效果及作用机制。方法 KM小鼠60只,随机分为空白组、模型组、皮肤康洗液组、瑶浴清消方高、中、低剂量组,每组各10只。建立耳部湿疹模型并同时外用给药干预治疗,观察小鼠耳廓皮肤损伤程度、肿胀度、HE染色病理切片以及小鼠血清中TNF-α、IFN-γ、IL-4的变化。结果 瑶浴清消方各组与模型组相比小鼠耳廓急性湿疹症状有所缓解,体现在耳廓皮肤损伤评分、肿胀度的降低和HE染色病理切片组织内炎症细胞的减少,以及TNF-α、IL-4含量表达的降低,IFN-γ含量表达升高,以上差异均有统计学意义(P<0.01),其中瑶浴清消方中剂量组干预效果最佳。结论 瑶浴清消方能有效缓解急性湿疹造成的小鼠耳廓皮肤损伤及肿胀度,减轻湿疹急性期炎症症状,其作用机制或是通过降低TNF-α、IL-4表达含量,升高IFN-γ表达含量,从而恢复Th1/Th2细胞平衡状态,达到治疗急性湿疹的目的。
Abstract:
Objective To investigate the intervention effect and mechanism of Yaoyu Qingxiao Formula(YYQXF)on acute eczema model of mice.Methods Sixty KM mice were randomly divided into blank group,model group,Pifukang Xiye group,high-dose YYQXF group,medium-dose YYQXF group,and low-dose YYQXF group,with 10 mice in each group.The model of ear eczema was established,and in the meantime the drug intervention was given.The degree of skin damage of auricle,auricle swelling,HE-stained pathological sections and the expression of TNF-α,IFN- γ,and IL-4 in serum of mice were observed.Results Compared with the model group,the symptoms of acute eczema on the auricle of mice in YYQXF groups were relieved,which was suggested in the reduction in skin damage score of auricle and auricle swelling degree,the decrease of inflammatory cells in HE-stained pathological sections,as well as reduced expression of TNF-α and IL-4,and increased expression of IFN- γ(P< 0.01).The intervention effect of medium-dose YYQXF group was the best.Conclusion YYQXF could effectively alleviate the skin damage of mice ear caused by acute eczema,reduce auricle swelling in mice,and alleviate inflammatory symptoms in mice with eczema at acute stage.Its mechanismcan restore the balance of Th1/Th2 cells, by decreasing the expression of TNF-α and IL-4 and increasing the expression of IFN- γ, thereby achieve the purpose of treating acute eczema.
