摘要:
目的 观察气压喷液仪喷射不同浓度复方甘草酸苷注射液(CG)对特应性皮炎(AD)样小鼠模型的影响。方法 用2,4-二硝基氯苯对雌性BABL/c小鼠进行AD造模,结合临床表现、无创检测及病理特征判断是否符合特应性皮炎改变; 将造模成功后的小鼠随机分为模型组、40%CG组、80%CG组、100%CG组和阳性对照组,每组6只。通过气压喷液仪进行给药,分别在给药第0、1、3、7天后对小鼠进行表皮失水(TEWL)检测、皮损拍照,给药7 d后小鼠摘眼球取血,ELISA检测血清总IgE水平; Western blot测定各组小鼠水通道蛋白3(AQP3)的蛋白表达水平; 正常对照组小鼠,常规饲养,不做任何处理。结果 造模后小鼠背部皮肤可见水肿、红斑、脱屑、结痂等炎症反应,组织病理示:角化过度、表皮层增厚、真皮层有淋巴细胞为主的炎性细胞浸润; 模型组小鼠TEWL值(75.3±5.43)g/(m2·h)、血清总IgE(12.64±1.14)pg/mL与正常对照组(11.76±0.49)g/(m2·h),(2.22±0.09)pg/mL相比明显增高,且差异具有统计学意义(P<0.05),综合以上结果表明在BABL/c小鼠上成功建立了 AD 模型; 经复方甘草酸苷注射液治疗后,小鼠背部皮肤皮损基本消退,症状好转; 小鼠TEWL值、血清总IgE水平及AQP3蛋白表达水平均低于模型组,差异具有统计学意义(P<0.05),综合分析3种不同给药浓度,其疗效存在差异,40%CG组<80%CG组<100%CG组,差异具有统计学意义(P<0.05)。结论 利用气压喷液仪喷射不同浓度的复方甘草酸苷注射液,可直接将药物导入特应性皮炎样小鼠模型的皮损处,在抗炎、抗过敏和类激素样作用的同时,可修复皮肤屏障功能,随着给药浓度的增加,临床疗效更显著。
Abstract:
Objective To observe the effect of spraying different concentrations of compound glycyrrhizin injection on atopic dermatitis-like mouse model by pneumatic sprayer.Methods 2,4-dinitrochlorobenzen(DNCB)was used to establish AD model of female BABL/c mouse,and clinical manifestations,non-invasive detection and pathological characteristics were combined to determine whether they were consistent with the changes of atopic dermatitis.After successful modeling,the mice were randomly divided into model group,40%CG group,80% CG group,100% CG group,and positive control group,with six mice in each group.The administration was carried out by a pneumatic sprayer.The mice were tested for transepidermal water loss(TEWL)and skin lesions were photographed at 0,1,3,and 7 d after administration.At 7 d after administration,the eyeballs of mice were harvested and blood was collected.The total serum IgE level was detected by ELISA,and the protein expression level of aquaporin 3(AQP3)in each group was measured by Western blot.Mice in the normal control group were raised routinely,without any treatment.Results After modeling,inflammatory reactions such as edema,erythema,desquamation,and scab were seen on the back skin of the mouse.Histopathology showed hyperkeratosis,thickening of the epidermis,and inflammatory cell infiltration mainly of lymphocytes in the dermis.TEWL value(75.3±5.43)g/(m2·h),and serum total IgE(12.64±1.14)pg/mL in model group were significantly higher than those(11.76±0.49)g/(m2·h)and(2.22±0.09)pg/mL,respectively in the normal control group,suggesting significant difference(P<0.05).The above results indicated that the AD model was successfully established on BABL/c mouse.After treatment with compound glycyrrhizin injection,the skin lesions on the back of the mouse practically subsided and the symptoms improved.The TEWL value,serum total IgE level,and AQP3 protein expression level were lower in treatment groups than in the model group,and the difference was statistically significant(P<0.05).Comprehensive analysis of three different administration concentrations showed that the efficacy was different,which was the lowest in 40%CG Group,followed by 80%CG group and 100%CG group,with significant difference(P<0.05).Conclusion By spraying different concentrations of compound glycyrrhizin injection with a pneumatic sprayer,the drug can be directly introduced into the skin lesions of atopic dermatitis-like mouse models,producing anti-inflammatory,anti-allergic and hormone-like effects.In addition,it can repair the skin barrier function,and the clinical effect is more significant with the increase in the concentration of compound glycyrrhizin injection.
