摘要:
目的 探讨红光对毛乳头细胞增殖和分泌血管内皮生长因子(VEGF)及肝细胞生长因子(HGF)的影响。方法 运用四甲基偶氮唑蓝(MTT)法检测不同剂量(0.5、1、2、4 J/cm2)红光(波长为635 nm)对毛乳头细胞增殖水平的影响。采用蛋白免疫印迹法(Western blot)和实时荧光定量PCR法(RT-PCR)检测不同剂量红光对毛乳头细胞分泌VEGF、HGF的影响。结果 红光能显著促进毛乳头细胞增殖,且照光剂量为1 J/cm2为最优剂量; Western blot和RT-PCR结果也显示,红光可上调VEGF、HGF表达水平,且照光剂量为1 J/cm2时,升高水平最为显著。结论 红光促进毛乳头细胞增殖的同时,可通过上调VEGF、HGF表达水平,从而促进毛发生长。
Abstract:
Objective To investigate the effect of red light on the proliferation of human dermal papilla cells(DPCs)and secretion of vascular endothelial growth factor(VEGF)and hepatocyte growth factor(HGF).Methods The effect of different doses(0.5,1,2,4 J/cm2)of red light(with a wavelength of 635 nm)on the proliferation of DPCs was detected by methyl thiazolyl tetrazolium(MTT).The effect of different doses of red light on the secretion of VEGF and HGF from DPCs was measured by western blot and real-time PCR(RT-PCR).Results The proliferation of DPCs treated with red light was increased,and 1 J/cm2was the optimal dose.The results of Western blot and RT-PCR showed that the expression levels of VEGF and HGF were enhanced after treated with red light.The highest level of expression was detected after treatment with red light with 1 J/cm2.Conclusion The red light could promote the proliferation of DPCs,and stimulate the hair growth by up-regulating the VEGF and HGF expression.
