目的 检测早老蛋白-1(presenilin1,PS1)在脂溢性角化病(seborrheic keratosis,SK)皮损及邻近正常皮肤组织(paired adjacent normal skin tissue,PANST)的表达,分析不同病理类型间的差异,探讨PS1与SK形成的相关性。方法 收集贵州医科大学附属医院皮肤科2017年9月-2020年1月SK患者41例,SK皮损及邻近正常皮肤组织行石蜡包埋及病理切片,HE染色后光学显微镜下行病理学诊断及分型。免疫组织化学及实时荧光定量PCR分别检测PS1蛋白及mRNA在SK皮损及邻近正常皮肤组织中的表达。蛋白印迹检测SK皮损及邻近正常皮肤组织中淀粉样前体蛋白(amyloid precursor protein,APP)、PS1的表达,并比较它们的差异。结果 41例SK的病理分型分别为角化过度型、腺样型、棘层增厚型、克隆型、激惹型及扁平型。免疫组织化学显示,PS1在SK皮损中的表达高于邻近正常皮肤组织,且表达于SK皮损组织的全层,邻近正常皮肤组织仅表达于表皮基底层细胞。PS1的mRNA水平在SK皮损中明显低于邻近正常皮肤组织,差异有统计学意义(P<0.001); 棘层增厚型中皮损处PS1 mRNA水平同样低于邻近正常皮肤组织,差异有统计学意义(P<0.05); 各病理类型间PS1表达差异无统计学意义。APP、PS1蛋白水平在SK皮损中高于邻近正常皮肤组织。结论 PS1在SK皮损及邻近正常皮肤组织,尤其在棘层增厚型中的差异表达,可能会为未来筛选老化标记及研究SK发病机制提供新的思路。
Abstract:
Objective To detect the expression of presenilin-1(PS1)in lesion tissue of seborrheic keratosis(SK)and paired adjacent normal skin tissue(PANST),as well as in different pathological types of SK,to explore the correlation between PS1 and SK formation.Methods Forty-one patients with SK treated in the Department of Dermatology,the Affiliated Hospital of Guizhou Medical University from September 2017 to January 2020 were selected.Paraffin embedded and pathological sections were conducted in SK lesions and PANST.Pathological diagnosis and classification were performed by optical microscope after HE staining.Immunohistochemistry and quantitative real-time PCR were used to determine the protein and mRNA expression of PS1 in SK lesions and PANST.The expression levels of APP and PS1 in SK lesions and PANST were detected by Western blot and compared.Results Of all 41 patients,SK specimens were classified into hyperkeratotic,acanthotic,adenoid,clonal,irritated and flat pathological types.Immunohistochemical staining showed that the expression of PS1 in SK lesions was higher than that in PANST,and it was expressed in the whole layer of SK lesions,while only in basal cells of epidermis in PANST.The mRNA level of PS1 in SK lesions was significantly lower than that in PANST(P<0.001),and the mRNA level of PS1 in the skin lesions of acanthotic type was also lower than that of PANST(P<0.05),but there was no significant difference in the expression of PS1 among the pathological types.By contrast,the protein level of APP and PS1 in SK lesions was higher than that in PANST.Conclusion The differential expression of PS1 in SK lesions and PANST,especially in acanthotic type,may provide a new idea for screening aging markers and studying the pathogenesis of SK in the future.
