摘要:
目的 探讨复方青黛胶囊在银屑病微环境下对HaCaT细胞IL-2、IL-6、IL-8、IL-18、IFN-γ表达水平的影响,初步阐明复方青黛胶囊治疗银屑病的可能机制。方法 在银屑病微环境下观察复方青黛胶囊对HaCaT细胞IL-2、IL-6、IL-8、IL-18、IFN-γ表达水平的影响,并设空白对照和阿维A阳性对照。将HaCaT细胞分为4组,对照组(C组)不加任何干预; 模型组(M组):采用M5(IL-1a、IL-17A、IL-22、抑癌蛋白M和TNF-α)模拟银屑病炎性微环境造模; 复方青黛胶囊组(MF组)在模型组基础上加10%复方青黛胶囊培养72 h; 阿维A组(MA组)在模型组基础上+10 μmol/L阿维A培养72 h。采用ELISA方法检查各组细胞培养上清中IL-2、IL-6、IL-8、IL-18、IFN-γ的水平。结果 M组与C组相比,IL-6、IL-8、IL-2、IFN-γ、IL-18水平显著升高,差异均有统计学意义(P均<0.05); MF组与M组相比IL-6、IL-8、IL-2、IFN-γ、IL-18水平显著降低,差异均有统计学意义(P均<0.05); MA组与M组相比IL-6、IL-2、IFN-γ、IL-18水平显著降低,差异均有统计学意义(P均<0.05),两组IL-8水平比较差异无统计学意义(P>0.05); MF组与MA组比较IL-6、IL-8、IFN-γ水平差异均有统计学意义(P均<0.05),两组IL-2、IL-18水平差异无统计学意义(P均>0.05)。结论 复方青黛胶囊可能通过降低IL-6、IL-8、IL-2、IFN-γ、IL-18等银屑病相关炎性因子的水平而发挥治疗银屑病的作用。
Abstract:
Objective To investigate the effect of compound Qingdai capsule on the expression levels of IL-2, IL-6, IL-8, IL-18 and IFN-γ in HaCaT cells under the microenvironment of psoriasis, and to preliminarily clarify the possible mechanism of compound Qingdai capsule in the treatment of psoriasis.Methods Cultured HaCaT cells were divided into 4 groups.The control group(C)was treated without any intervention.Model group(M)were cultured with IL-1A, IL-17A, IL-22, tumor suppressor protein M and TNF-γ to simulate the inflammatory microenvironment of psoriasis.Compound Qingdai capsule group(MF)was cultured with 10% compound Qingdai capsule for 72 h on the basis of model group.Acitretin group(MA)was cultured for 72 h with 10 mol/L acitretin on the basis of model group.The levels of IL-2, IL-6, IL-8, IL-18 and IFN-γ in the supernatant of cell culture in each group were examined by ELISA.Results The levels of IL-6, IL-8, IL-2, IFN-γ and IL-18 in M group were significantly higher than those in C group, and the differences were statistically significant(all P<0.05).The levels of IL-6, IL-8, IL-2, IFN-γ and IL-18 in MF group were significantly lower than those in M group, and the differences were statistically significant(all P<0.05).The levels of IL-6, IL-2, IFN-γ and IL-18 in MA group were significantly lower than those in M group with statistically significant differences(all P<0.05), while the levels of IL-8 in MA group and M group were not significantly different(P>0.05).There were statistically significant differences in IL-6, IL-8 and IFN-γ levels between MF and MA group(all P<0.05), while no statistically significant differences in IL-2 and IL-18 levels between MF and MA group(all P>0.05).Conclusion Compound Qingdai capsule may play a role in the treatment of psoriasis by reducing psoria-related inflammatory factors such as IL-6, IL-8, IL-2, IFN-γ and IL-18.
