摘要:
目的 探讨龙血竭凝胶对兔耳痤疮模型TNF-α、IL-6和MFGE8的作用效果。方法 24只新西兰家兔随机分为空白组4只和模型组20只,模型组以煤焦油涂抹和痤疮丙酸杆菌接种制备兔耳痤疮模型后,随机分为模型组、阳性药组、龙血竭凝胶高、中、低剂量组[根据对痤疮丙酸杆菌(P.acne)、金黄色葡萄球菌(S.aureus)、表皮葡萄球菌(S.epidermidis)的MIC确定],每组4只,涂抹给药,空白组和模型组给予空白凝胶基质,持续14 d。观察兔耳厚度及病理组织结构,ELISA法检测血清中TNF-α、IL-6含量,Western blot检测耳片组织中MFGE8表达。结果 龙血竭凝胶对痤疮丙酸杆菌的MIC为62.5 mg/mL,对金黄色葡萄球菌的MIC为125 mg/mL,对表皮葡萄球菌的MIC为31.25 mg/mL。故以31.25、62.5、125 mg/mL为低、中、高剂量给药,14 d后发现,中、高剂量龙血竭凝胶可不同程度改善大鼠耳廓痤疮模表皮角化、减轻炎性细胞浸润及脓性丘疹。阳性对照组和龙血竭凝胶高剂量组兔耳厚度与模型相比均有降低(P<0.05),兔血清中TNF-α、IL-6水平与模型组相比均有降低(P<0.01),空白组、阳性对照组和高剂量组耳片组织中MFGE8表达高于模型组(P<0.01)。结论 外用涂擦龙血竭125 mg/mL、62.5 mg/mL凝胶对兔耳痤疮有改善作用,这可能与其能够降低其血清中TNF-α、IL-6水平,恢复痤疮模型兔耳组织中MFGE8的表达有关。
Abstract:
Objective Study the effect of the resin from Dracaena cochinchinensis(RDC)gel on the TNF-α,IL-6 and the Milk Fat Globule-EGF-factor 8(MFGE8)in acne model of rabbits' serum and ear skin. Methods Twenty-four New Zealand rabbits were randomly divided into blank group with 4 rabbits and model group with 20 rabbits. Model group was built rabbit ear acne model with coal tar daub and acne propionic acid bacillus. After the success of the acne model,the rabbits divided into 5 groups including model group,positive control group(Adapalene gel)and the low,medium and high dose RDC gel groups,with 4 rabbits for each group. The blank group was given a blank gel matrix. Daubing lasted 14 days for once a day. Observed the appearance and the pathological organization structure of rabbit ears. The thickness was measured with Vernier caliper. The level of TNF-α,IL-6 in serum was detected by ELISA,and MFGE8 expression in the rabbits' ear skin tissue was analyzed by Western blot. Results 31.25,62.5,125 mg/mL RDC gel was tested in the experimental groups,which was based on the MIC result to S.epidermidis,P.acne,S.aureus and After 14 days,the medium and high dose RDC gel relieved auricle acne epidermal keratinization,reduced inflammatory cell infiltration and purulent papules. The ear thickness of the positive control group and RDC gel high dose group were lower than the model of group(P<0.05),and the rabbit serum TNF-a,IL-6 levels are also lower than the model group(P<0.01). Compared with the model group,the MFGE8 expression in ears for the blank group,positive control group and high dose group was higher(P<0.01),and there were no significant differences in the three groups. Conclusion 125 mg/mL,62.5 mg/mL RDC gel were effective in the treatment of acne. It may be not only related to the inhibition and reduction of TNF-α and IL-6 production,but also enhanced MFGE8 expression in acne model rabbits' ears.
