摘要:
目的 探讨A型肉毒素是否通过MAPK信号通路下调UVB诱导的HaCaT细胞炎症因子的表达。方法 培养HaCaT细胞,分为4组:对照组(未做任何处理); UVB组(80 mJ/cm2); UVB+A型肉毒素组(80 mJ/cm2+5 IU/mL); A型肉毒素组(5 IU/mL)。采用酶联免疫吸附试验检测HaCaT细胞培养液中的IL-6及IL-8细胞炎症因子的表达情况。通过蛋白免疫印迹法测定细胞内MAPK相关通路蛋白的表达水平。结果 与UVB组相比,UVB+A型肉毒素组IL-6及IL-8细胞炎性因子释放减少。与对照组相比,UVB组p-ERK、p-JNK表达水平增加。UVB+A型肉毒素组与UVB组比较,p-ERK、p-JNK表达水平下降,而p-AKTSer473及p-P38无明显改变。结论 A型肉毒素可能通过p-ERK、p-JNK信号通路下调UVB诱导的HaCaT细胞炎症因子的表达。
Abstract:
Objective To investigate whether botulinum toxin type A(BoNTA)down-regulates UVB-induced expression of inflammatory cytokines in HaCaT cells via MAPK signaling pathway.Methods HaCaT cells were cultured and divided into 4 groups:control group(without any treatment),UVB group(80 mJ/cm2),UVB+BoNTA group(80 mJ/cm2+5 IU/mL),BoNTA group(5 IU/mL).The expression levels of IL-6 and IL-8 were measured by enzyme linked immunosorbent assay.The expression levels of MAPK pathway proteins were determined by Western blot.Results Compared with the UVB group,the UVB+BoNTA group significantly reduced levels of IL-6 and IL-8.In comparison to the controls,UVB alone increased the expression levels of p-ERK and p-JNK.But UVB+BoNTA treatment decreased expression levels of p-ERK and p-JNK without significant changes in p-AKTSer473and p-P38 in comparsion to UVB alone.Conclusion Botulinum toxin type A down-regulates UVB-induced expression of inflammatory cytokines in HaCaT cells probably via p-ERK,p-JNK signaling pathways.
