摘要:
目的 探究芦荟提取物芦荟多糖(AP)对特应性皮炎小鼠的保护作用以及对免疫球蛋白E(IgE)、白介素-17(IL-17)及核因子κB(NF-κB)通路的影响。方法 将小鼠分为对照组(control)、模型组(model)、AP低、中、高剂量组,阳性对照组(波尼松龙,prednisolone),对小鼠背部受损皮肤进行评分,HE染色观察皮肤组织病理损伤; 测定小鼠脾脏、体重、脾脏指数变化; 酶联免疫法(ELISA)检测血清IgA、IgM、IgE水平; 流式细胞术检测脾脏淋巴液中Th17细胞含量; 蛋白免疫印迹(WB)、免疫组织化学法检测受损皮肤组织IL-17、NF-κB蛋白表达。结果 与对照组相比,模型组受损皮肤评分显著升高; 与模型组相比,AP低、中、高剂量组、波尼松龙组受损皮肤评分显著降低(P<0.05)。病理切片提示模型组小鼠背部皮肤角化严重,出现大量炎性细胞浸润; AP低、中、高剂量组、波尼松龙组皮肤组织得到明显缓解。与对照组相比,模型组体重、脾脏重量、脾脏指数降低,IgA、IgE水平显著升高,IgG水平显著降低,Th17细胞比例显著升高,IL-17、NF-κB蛋白表达显著升高(P<0.05); 与模型组相比,AP低、中、高剂量组、波尼松龙组体重、脾脏重量、脾脏指数显著升高,IgA、IgE水平显著降低,Th17细胞比例显著降低,IL-17、NF-κB蛋白表达显著降低(P<0.05)。结论 芦荟多糖对特应性皮炎小鼠皮肤具有一定的保护作用,其机制与抑制Th17细胞以及NF-κB通路有关。
Abstract:
Objective To investigate the protective effect of aloe polysaccharide(AP)on atopic dermatitis mice model and its influences on immunoglobulin E(IgE),interleukin-17(IL-17)and nuclear factor-κB(NF-κB)pathway.Methods Mice were divided into control group,model group,AP low,medium and high dose groups,positive control group(prednisolone).The impaired skin on the back of mice was scored,HE staining was used to observe pathological changes of skin tissues.The spleen weight,body weight and spleen index of mice were measured.Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of IgA,IgM and IgE in serum; the content of Th17 cells in spleen lymph was detected by flow cytometry; Western blot(WB)and immunohistochemistry were used to detect the expressions of IL-17 and NF-κB protein in impaired skin.Results Compared with control group,the impaired skin score of model group increased significantly; compared with model group,the impaired skin scores of AP low,middle and high dose groups and prednisolone group were significantly decreased(P<0.05).Pathological section showed severe keratosis on the back skin of mice in model group,and a large number of inflammatory cell infiltration occurred; the skin tissues of AP low,middle and high dose groups and prednisolone group were significantly relieved.Compared with control group,body weight,spleen weight and spleen index decreased in model group,the levels of IgA and IgE increased significantly,the level of IgG decreased significantly,the proportion of Th17 cells increased significantly,and the protein expressions of IL-17 and NF-κB increased significantly(P<0.05).Compared with model group,the body weight,spleen weight and spleen index of AP low,middle and high dose groups and prednisolone group were significantly increased,the levels of IgA and IgE decreased significantly,the proportion of Th17 cells decreased significantly,and the expressions of IL-17 and NF-κB were significantly decreased(P<0.05).Conclusion Aloe polysaccharide has protective effect on the skin of mice with atopic dermatitis.The mechanism is related to the inhibition of Th17 cells and NF-κB pathway.
