摘要:
目的探讨女性SLE患者骨桥蛋白(OPN)基因rs2853750及rs6811536突变差异与发病风险及不同并发症的相关性。方法随机选取2013-2015年经广西右江民族医学院附属医院确诊的200例女性SLE患者作为病例组,并选取同期在本院进行体检的200名同性别健康人作为对照组,采用单碱基延伸PCR技术和DNA测序法检测OPN基因多态性,对比SLE组与健康组、SLE组与SLE并发症组的OPN基因rs2853750和rs6811536位点的基因型和等位基因分布频率,并分析OPN基因单倍型频率。结果广西人群中SLE组和健康对照组OPN rs2853750和rs6811536位点的基因型和等位基因频率以及基因单倍型频率差异均无统计学意义,rs2853750基因型AG和等位基因(A/G)频率与SLE合并关节炎相关,发病风险差异具有统计学意义(P=0.035,0.04);rs6811536基因型和等位基因在伴并发症及不伴并发症组间发病风险差异无统计学意义(PSymbol~@@0.05)。结论广西女性OPN rs2853750和rs6811536基因多态性与SLE的发病风险无关,rs2853750可能是SLE女性患者并发关节炎的易感位点。
Abstract:
Objective To investigate the relationship between the Osteopontin gene rs2853750 and rs6811536 mutations, and the risk and complications in females with Systemic Lupus Erythematosus (SLE).MethodsA total of 200 females with SLE and 200 healthy females undergoing routine physical examination were enrolled from the Affiliated Hospital of Youjiang Medical University for Nationalities of Guangxi from 2013 to 2015.The Single-base extension PCR and DNA sequence assay were used to detect the OPN gene polymorphism.The genotype and allele distribution frequency of OPN gene rs2853750 and rs6811536 were compared between SLE and healthy controls, SLE and SLE with complications, in addition to analysis of the OPN gene haplotype frequency.ResultsThere were no differences in genotype, allele frequency and gene haplotype frequency of both OPN rs2853750 and rs6811536 loci between SLE and healthy controls in Guangxi population.However, rs2853750 genotype AG and allele (A/G) frequency, but not rs6811536, were associated with SLE complicated with arthritis and the risk of arthritis (P=0.035,0.04).ConclusionThe polymorphisms of OPN rs2853750 and rs6811536 genes in Guangxi women are not associated with the risk of developing SLE, and rs2853750 may be a susceptibility site for female SLE patients with arthritis.
