摘要:
目的 观察研究特异性免疫治疗对BALB/c小鼠特应性皮炎模型的疗效。方法 雌性的SPF级纯系BALB/c小鼠36只,随机分为空白对照组、卵清蛋白激发组、丙酮激发组三组,每组12只。应用尘螨溶液诱导小鼠形成过敏体质,再分别用0.5%的丙酮和卵清蛋白诱导小鼠产生特应性皮炎,对照组选择等量PBS致敏处理。造模成功后,每组随机选择6只小鼠取血,酶联免疫吸附法(Elisa)检测血清总IgE水平变化。取血完毕后取相应小鼠皮肤进行病理切片。剩余小鼠应用阿罗格脱敏试剂进行特异性免疫治疗(specific immunotherapy,SIT)1周,对照组未予任何处理,取血检测血清IgE水平,相应皮损进行病理组织切片。结果 接触卵清蛋白及丙酮的小鼠皮肤局部呈现表皮增生、细胞浸润等炎症性改变,与正常对照组相比较,临床表现及病理符合特应性皮炎改变; 实验组小鼠血清总IgE(卵清组与丙酮组数值:3 479.899 ±758.945pg /mL、3 809.807 ±532.886pg /mL)与对照组数值(1 148.970 ±131.648pg/mL)相比明显升高,差异具有统计学意义(P<0.05)。经特异性免疫治疗一周后,实验组小鼠血清总IgE水平与前对照降低,表皮增厚及炎性渗出症状改善。结论 卵清蛋白与丙酮在尘螨致敏小鼠表皮上均可产生类似人类特应性皮炎的临床及病理改变,为AD发病机制及治疗的研究提供一个较理想的动物模型。特异性免疫治疗对人类特应性皮炎炎症的转归有一定效果。
Abstract:
Objective To study the effect of specific immunotherapy in atopic dermatitis(AD)mice model. Methods Thirty-six BALB/c mice were randomly divided into three groups,which were control group, acetone(DNCB)group and ovalbumin(OVA)group.The atopic dermatitis mice were established by sensitization with dust mite allergy solution, and then were treated with 0.5% DNCB or ovalbumin on the skin, while the mice of control group were treated with PBS. After 6 weeks, Enzyme-linked immunosorbent assay(Elisa)was performed to detect the level of the IgE in serum. The severity of skin lesions was examined by macroscopically and histologically methods. After that we used the specific immunotherapy(SIT)to treat the mice for one week.The same methods were used to observe the changes, respectively.Results The ovalbumin and acetone induced clinical symptoms and the histopathological changes such as epidermal hyperplasia and infiltration of mast cells and eosinophils in the dermis. The total IgE of experimental groups serum(OVA group and DNCB group and the control group numerical values:3479.899±758.945pg/mL.3809.807±532.886pg/mLvs.1148.970±131.648pg/mL)significantly increased, the difference was statistically significant(P<0.05). After the specific immunotherapy treatment for one week,Enzyme-linked immunosorbent analysis confirmed that the groups of the experimental groups had the lower level of the IgE,compared with before. The clinical symptoms and the histopathological changes were improved after the treatment.Conclusion The above results suggest that ovalbumin and the acetone act on the skin which was sensitized with dust mites can induce atopic dermatitis in BALB/c mice. The specific immunotherapy may be useful for the treatment of AD.
