摘要:
目的 研究蒺藜防白丸对黑素细胞(melanocytes,MC)的形态、增殖、酪氨酸酶活性及黑素合成的影响。方法 采用倒置显微镜观察细胞形态,四甲基偶氮唑蓝(MTT)法测定蒺藜防白丸对黑素细胞增殖的影响,多巴氧化法测定酪氨酸酶活性,NaOH法测定黑素含量。结果 随着蒺藜防白丸浓度的升高,黑素细胞树突变粗短,且数量减少; 低浓度(0.001~0.5mg/mL)的蒺藜防白丸对黑素细胞增殖影响不显著(P>0.05),高浓度时(1mg/mL和5mg/mL)抑制作用明显(P<0.05); 低浓度的蒺藜防白丸可以提高酪氨酸酶活性,其中0.05mg/mL时酪氨酸酶活性最高(P<0.05),然而当蒺藜防白丸浓度继续增大,则会对酪氨酸酶活性产生抑制作用; 蒺藜防白丸对黑素细胞黑素合成也有类似的影响。结论 蒺藜防白丸高浓度时抑制黑素细胞的增殖、酪氨酸酶活性及黑素合成; 低浓度时促进酪氨酸酶活性及黑素合成。
Abstract:
Objective To evaluate the effect of Jilifangbaiwan on morphology, proliferation, tyrosinase activity and melanin synthesis in cultured normal human melanocytes. Methods Cell morphology was observed with inverted microscope. Cell proliferation, tyrosinase activity and melanin content of cultured normal human melanocytes were measured by MTT, dopa-oxidase and NaOH method, respectively. Results The dendrites of the melanocytes became shorter and less as the concentration of Jilifangbaiwan was increased. Although Jilifangbaiwan at low concentration(0.001~0.5mg/mL)did not affect melanocyte proliferation, it significantly inhibited melanocyte proliferation at higher concentrations(1mg/mL and 5mg/mL)(P<0.05). The Jilifangbaiwan at low concentration increased the activity of tyrosinase with peak activity at the concentration of 0.05mg/mL(P<0.05). In contrast, Jilifangbaiwan at higher concentration inhibited tyrosinase. The effect of Jilifangbaiwan on melanin synthesis was in the similar pattern to that on tyrosinase activity.Conclusion Jilifangbaiwan at high concentration inhibits melanocyte proliferation, tyrosinase activity and melanin synthesis, while at low concentration it increases tyrosinase activity and stimulates melanin synthesis.
